Co-amoxiclav is an antibacterial combination of two active ingredients: Amoxicillin, an antibiotic, and Clavulanic Acid, a beta-lactamase inhibitor.

• Amoxicillin: Exhibits a broad spectrum of bactericidal activity against a variety of Gram-positive and Gram-negative microorganisms. However, it is vulnerable to degradation by beta-lactamases, enzymes produced by certain bacteria that render many penicillins ineffective.
• Clavulanic Acid: Inactivates a wide range of beta-lactamase enzymes that are commonly found in microorganisms resistant to penicillins and cephalosporins. By inhibiting these enzymes, Clavulanic Acid protects Amoxicillin from degradation, thereby extending its antibiotic spectrum to cover a wider range of bacteria.

The pharmacokinetics of Amoxicillin and Clavulanic Acid in Co-amoxiclav are well-matched, ensuring simultaneous peak serum levels.

• Absorption: Peak serum levels of both components are achieved approximately one hour after oral administration. Absorption is optimized when the medication is taken at the start of a meal.
• Serum Binding: Both Clavulanate and Amoxicillin have low serum protein binding, with about 70% remaining free in the serum.
• Dosage Correlation: Doubling the dosage of Co-amoxiclav approximately doubles the serum levels of both Amoxicillin and Clavulanic Acid.
